Antabuse (disulfiram; drug used for treating alcohol dependence) is a medication approved for use in treatment of alcohol use disorder. However, it is not available over-the-counter (OTC) in the USA and Europe. Antabuse has been found to be effective in reducing alcohol withdrawal symptoms, but it is still an experimental therapy. It is unclear how Antabuse works to treat alcohol dependence and its pharmacological effects. The aim of this study was to compare the pharmacokinetics of the new drug in healthy volunteers and to compare it with the established treatment of alcohol dependence.
A total of 12 healthy volunteers (3 male, 7 female; age range, 18–80 years, average body weight, 9.6 kg) were enrolled into the study. The study was performed in two phases: (1) a fast-track phase of the study and (2) a maintenance phase of the study. The primary outcome was the pharmacokinetic parameter estimation. The study was conducted in the second phase, in which participants were recruited from the public hospital. The study was approved by the institutional Ethics Committee of the First Affiliated Hospital of Guangxi Medical University (approval number: 812-09), and was carried out in accordance with the Declaration of Helsinki, Good Clinical Practice and the International Conference on Harmonisation guidelines. The patients were randomly assigned into two groups: a placebo group, which was given Antabuse tablets daily for three days, and a group treated with an alcohol-dependence-alcohol (ADOE) tablet for three days. The patients were given the same dose of ADOE tablets, but the number of tablets consumed during the study period. The patients were followed up for 3 months after treatment and at the end of the study, the mean abstinence rate was approximately 12.2%, which was maintained after two and six months. The pharmacokinetic parameters were calculated using the following formula:Ki=max/t×AC. The study was performed in a double-blinded manner, so as to be safe, both groups received the same dose of ADOE tablets.

The groups were administered the same dose of ADOE tablets, but the number of tablets consumed during the study period was not changed. The subjects in the placebo group were given the same dose of ADOE tablets and ADOE tablets for three days. The study was performed in a double-blinded manner, so as to be safe, both groups received the same dose of ADOE tablets and ADOE tablets for three days. The volunteers were followed up for 3 months after treatment and at the end of the study, the mean abstinence rate was approximately 12.2% in the placebo group and approximately 12.1% in the ADOE group.
